Del Puerto H.L., Vasconcelos A.C., Moro L., Alves F., Braz G.F. & Martins A.S. 2010. Canine distemper virus detection in asymptomatic and non vaccinated dogs. Pesquisa Veterinária Brasileira 30(2):139-144. Departamento de Patologia Geral, Instituto de Ciências Biológicas, Universidade Federal de Minas Gerais, Campus Pampulha, Belo Horizonte, MG 31270-901, Brazil. E-mail: [email protected] A quantitative real time polymerase chain reaction (PCR) revealed canine distemper virus presence in peripheral blood samples from asymptomatic and non vaccinated dogs. Samples from eleven domestic dogs with no signs of canine distemper and not vaccinated at the month of collection were used. Canine distemper virus vaccine samples in VERO cells were used as positive controls. RNA was isolated with Trizol®, and treated with a TURBO DNA-free kit. Primers were designed for canine distemper virus nucleocapsid protein coding region fragment amplification (84 bp). Canine β-actin (93 bp) was utilized as the endogenous control for normalization. Quantitative results of real time PCR generated by ABI Prism 7000 SDS Software showed that 54.5% of dogs with asymptomatic canine distemper were positive for canine distemper virus. Dissociation curves confirmed the specificity of the real time PCR fragments. This technique could detect even a few copies of viral RNA and identificate subclinically infected dogs providing accurate diagnosis of this disease at an early stage.

Molecular and Immunological Investigation of Canine Distemper Virus (CDV) and Its Co- Infection with Canine Parainfluenza Virus Type 2

Background and Objective: Canine distemper (CD) is a highly transmissible serious disease of carnivores. Distemper virus has immunosuppression effects, which, in turn, could lead to opportunistic infections. The present study was performed to detect CDV by the genomic and immunological methods and investigate its co- infection with canine parainfluenza virus type 2 (CPiV-2). Materials and Meth...

Detection and differentiation of wild-type and vaccine strains of canine distemper virus by a duplex reverse transcription polymerase chain reaction

Canine distemper virus (CDV) is the cause of canine distemper (CD) which is a severe and highly contagious disease in dogs. In the present study, a duplex reverse transcription polymerase chain reaction (RT-PCR) method was developed for the detection and differentiation of wild-type and vaccine strains of CDV. Four primers were designed to detect and discriminate the two viruses by generating 6...

Detection of Arctic and European cluster of canine distemper virus in north and center of Iran

Canine distemper virus (CDV) creates a very contagious viral multi-systemic canine distemper (CD) disease that affects most species of Carnivora order. The virus is genetically heterogeneous, particularly in section of the hemagglutinin (H) gene. Sequence analysis of the H gene can be useful to investigate distinction of various lineages related to geographical distribution and CDV molecular ep...

Molecular and Serological Detection of Canine Distemper Virus (CDV) in Rural Dogs, Iran

Background and Aims: Canine distemper (CD) is a deadly infectious disease of Canidae family. CD is a multi-systemic viral disease and is specified by wide range of clinical symptoms. The manifestations are not always indicative of CD, therefore a laboratory confirmation is necessary for suspected cases. Materials and Methods: Different clinical specimens of 19 CD suspected unvaccinated dogs wer...

Associations between biosecurity and outbreaks of canine distemper on Danish mink farms in 2012–2013

BACKGROUND During 8 months from July 2012 to February 2013, a major outbreak of canine distemper involving 64 mink farms occurred on the Danish peninsula of Jutland. The canine distemper outbreak was associated with exposure of farmed mink to infected wild carnivores and could represent a deficit in biosecurity on the mink farms. The aim of this study was to investigate the extent and associati...
